The successful candidate will be responsible for executing and managing an NIH funded project involving the genetic analysis of Anopheles mosquitoes using newly developed gene and enhancer-trapping technologies. This is a one-year appointment with the possibility of a second year depending on progress and performance. We recently developed functional Gal4-based gene- and enhancer-trapping technologies for the malaria mosquito Anopheles stephensi (O'Brochta et al, 2011; O'Brochta et al, 2012). These new forward genetic technologies provide new opportunities for exploring the physiological genetics of vectoral capacity. Two NIH-funded projects are available. The first project will use gene-trap technology to identify and analyze genes expressed specifically in the salivary glands of adult females. This project will provide unique opportunities to study the patterns of gene expression as it relates to salivary gland function under a variety of physiological conditions and will also provide an opportunity to evaluate hypomorphic and null mutations of identified genes on feeding and Plasmodium development. The second project will focus on the identification of hemocyte-specific enhancers. The Gal4-based enhancer-trap lines the will be created will initially be used to investigate the temporal and spatial dimensions of hematopoiesis during development and under a variety of physiological conditions including infection with Plasmodium, and eventually to genetically manipulate hemocyte populations and gene expression within hemocytes. The duties of this position will include designing and executing genetic crosses for the purposes of identifying genes and enhancers relevant to the vectoral capacity of Anopheles mosquitoes, molecular genetic analysis of genes and enhancers identified during genetic screens, supervising junior technical staff assigned to the project, preparing data for reporting and publication, presenting data to colleagues.
